Body of Abstract: The shape of the root system has a great influence on the growth and drought tolerance of crops. At present study, two japonica rice, Tainung 67 (TNG67) and Tainan 11 (TN11), an indica rice, Tainung Sen 14 (TNGS14), and the indica-japonica hybrid line, ATL382, were cultivated for 3 weeks with hydroponic solution supplemented different nitrogen, phosphorus and potassium nutrient levels. The seedling characters were further observed. The results showed that all four cultivars had similar growth trends at different growth stage and nutrition conditions. TNG67 and TN11 had significantly fewer root numbers and longer root length than TNGS14 and ATL382 especially at the first week. A set of Tetra-ARMs (tetra-primer amplification refractory mutation system) primers were designed from OsGRAS-like gene to distinguish single nucleotide polymorphism among these lines. It showed that TNG67 and TN11 belonged to C/C allele, while TNGS14 and ATL382 belonged to T/T allele at the 2282 nucleotide of OsGRAS-like gene. It belongs to synonymous substitution. This molecular marker can be applied to the screening of root morphology at seedling stage. Besides, OsGRAS-like gene expression were enhanced by phosphorus deficiency treatment and was strongly correlated to root length in rice seedling.
